  • 現今是以Alpha-fetoprotein(AFP)針對肝細胞癌的高危險族群做為肝癌篩檢的指標,約 70% 的肝癌患者AFP 會明顯上升;但由於AFP只有70%的檢測率,對於我們想要早期發現早期治療尚有難度,因此我們希望利用更多種的肝癌指標蛋白來做檢測,藉此可以提高肝癌檢測的靈敏度。
    在此研究中我們將Alpha-fetoprotein(AFP),Glypican3(GPC3),Des gamma carboxy prothrombin(DCP) 等抗體與磁性奈米粒子結合,形成具有磁性生物標記(magnetic bio – marker)之三種不同肝癌指標之磁性試劑(magnetic reagent);同時我們也開發出將AFP、GPC3、DCP三種抗體混合後在將混合抗體與磁性奈米粒子結合形成新的披覆多重抗體試劑(Multiple Coating);最後,再利用本實驗室所研發之磁減量檢測法(Immunomagnetic rednction ; IMR)以及顯微鏡以及ELISA等實驗器材,探討磁性試劑的特性,並與臨床檢測常用的酵素連結免疫吸附分析來進行比較;以及利用混合試劑量測混合標準抗原,探討其檢測標準值。
