本論文利用具有解酒以及護肝功能的漢方食材葛花以及枳椇子搭配辣木籽粉組合成草藥配方GLJ,探討GLJ對於肝臟細胞發炎與小鼠急性酒精性肝損傷的保護作用。我們採用實驗小鼠作為動物實驗模式,分成空白處理組(Sham)、只有給予草藥配方GLJ處理組(GLJ)、只有給予急性酒精處理組(ALC)、以及餵食草藥配方GLJ後再給予急性酒精處理組(GLJ+ALC)。利用小鼠平衡桿行走、滾輪跑步、懸吊測試,以及抽血檢測血液中天門冬胺基酸胺基轉移酶(GOT)、丙胺基酸胺基轉移酶(GTP)、以及乳酸脫氫酶(LDH)生化值。此外檢視小鼠肝臟組織,來探討草藥配方GLJ對於急性酒精處理小鼠的行為異常與肝功能保護作用。實驗結果顯示:ALC組小鼠相較於Sham與GLJ組小鼠的運動以及平衡的表現明顯差;而GLJ+ALC組小鼠相較於ALC組小鼠的運動以及平衡的表現則顯著改善許多。此外,ALC組小鼠相較於Sham與GLJ組小鼠血液內GOT、GTP、LDH的數值明顯較高,並且具有急性酒精性肝損傷的情形;而GLJ+ALC組小鼠相較於ALC組小鼠血液內GOT、GTP、LDH的數值則顯著降低,而急性酒精性肝損傷的情形也顯著改善。綜合上述結果,我們認為草藥配方GLJ處理對於急性酒精處理的肝細胞發炎以及小鼠的肝損傷應該具有保護的作用。由於葛花、枳椇子、辣木籽均屬藥食同源的草藥食材,因此,應該可以作為預防急性酒精肝損傷的營養補充品。
In this paper, we used the herbal ingredients of flowers of kudzuvine (Pueraria phaseoloides (Roxb.) Benth.), seeds of raisin tree (Hovenia acerba Lindl.), and together with seeds of moringa (Moringa oleifera Lam.), which have anti-alcoholic and liver-protecting functions, to form herbal formula GLJ. Then we explored the protective effect of GLJ on acute alcoholic liver injury in mice. We used experimental mice as the animal experiment model. Mice were divided into blank treatment group (Sham), herbal formula GLJ treatment group (GLJ), acute alcohol treatment group (ALC), and acute alcohol treatment after feeding GLJ group (GLJ+ALC). We further studied the protective effects of GLJ on the behavior and liver dysfunction of acute alcohol-treated mice. Behavior dysfunction was examined by using wheel running, balance pole walking, and suspension tests. Liver dysfunction was examined by detecting biochemical value of aspartate aminotransferase (GOT), alanine aminotransferase (GTP), and lactate dehydrogenase (LDH) in blood, and examining liver tissue. Our results showed that the performance of movement and balance of the ALC mice group was significantly worse than that of the Sham and GLJ mice groups; compared with the ALC mice group, the performance of movement and balance of the GLJ+ALC mice group was significantly improved. In addition, compared with the the Sham and GLJ mice groups, the values of GOT, GTP, and LDH in the blood of the ALC mice group were increased significantly, and acute alcoholic liver injury occurred; while compared with the ALC mice group, the values of GOT, GTP, and LDH in the blood of the GLJ+ALC mice group were significantly reduced, and the situation of acute alcoholic liver injury was also significantly improved. Based on the above results, we believe that the herbal formula GLJ treatment should have a protective effect on acute alcohol treated liver cell inflammation and liver injury in mice. Thus, we suggested that herbal formula GLJ can be used as nutritional supplements to prevent acute alcoholic liver injury because they are homology of medicine and food.
