在臨床診斷上約 60%~70% 的肝癌患者血清中Alpha-fetoprotein ( AFP ) 濃度較正常人來的高 ( AFP cut-off 值為 20 mg/ml ),適合作為篩檢肝癌的腫瘤指標蛋白 ( biomarker )。另外在臨床診斷上約50%~80%的大腸癌患者血清中Carcinoembryonic Antigen ( CEA ) 濃度較正常人來的高 ( CEA cut-off 值為 5 ng/ml ),適合作為篩檢大腸癌的 biomarker。然而現行普遍使用的免疫檢測如酵素連結免疫吸附分析 ( Enzyme-Link Immunosorbent Assay;ELISA ) 因其操作過程繁雜與易受待測樣品影響等種種問題,我們需要另尋一種更快速且準確的免疫檢測方法。
本實驗室已歷經使用Anti-CRP( C-reactive protein )與奈米磁性粒子結合,形成一具有磁性生物標記( magnetic biomarker ) 之磁性試劑( magnetic reagent )。再利用本實驗室所研發之磁減量檢測法( Immunomagnetic rednction ; IMR ),其靈敏度可達10-4 mg/ml,於是本研究中我們如法泡製將Anti-AFP 和Anti-CEA 分別與奈米磁性粒子結合成為AFP試劑和CEA試劑,並進行試劑長期穩定度觀察、干擾度測試,最後藉由IMR檢測人類血清檢體,以探討人類肝癌及大腸癌之檢驗特性。
