感染類志賀氏毒素會導致腸道出血性大腸炎,還會引起危險的溶血尿毒症候群,嚴重者更可能會造成腎臟的永久敗壞和中樞神經的傷害。而其感染途徑是藉由先結合到細胞膜表面,再行釋放毒素,所以若能抑制其結合到細胞膜表面,則可以抑制此毒素。類志賀氏毒素為AB5結構,A單位是毒性部分而B單位則是與細胞結合的部分。B單位是一個五聚體,每一個單體上有三個結合點,與細胞膜上的Pk抗原有特異性的結合力,這種多價牙基與多價受器的結合,稱為多價作用力(Multivalent interaction)。本論文包括I型類志賀式毒素B單位的純化以及利用金奈米粒子作為多價載體設計抑制劑,包含有不同大小的金奈米粒子,和不同鏈長的Pk抗原類似物包覆其上,利用表面電漿共振光譜儀測量I型類志賀式毒素B單位與Pk抗原類似物的動力學解離常數,研究金奈米粒子大小及Pk類似物鏈長改變後抑制效果的比較。
