C-反應蛋白(C-reactive Protein, CRP)是一種發炎反應期肝臟製造的敏感性產物,最近研究指出在心臟病或癌症病人血漿中的含量較高,可被視為是一項冠狀動脈的危險因子。因此,在臨床診斷化驗室裡微量CRP之敏感度、準確性及快速分析是急切地需要的。
目前醫療上對CRP的檢測大部分為光學方式,例如:免疫散射比濁法(immunosorbent nephelometry assay, INA)與酵素連結免疫吸附分析法(enzyme-linked immunosorbent assay, ELISA)。如果檢測的樣品帶有顏色,那麼在量測上的準確性將大打折扣。
於是在此研究裡,我們用來量測人體血清中感染因子C-反應蛋白的檢測儀器為磁性標記(magnetically labeled)之混頻交流磁導率計(mixed-frequency ac magnetosusceptometer),試劑為取自山羊的抗體(anti-CRP)與分散於水中的磁性奈米粒子(magnetic nanopartical)結合而成的磁性流體。實驗中我們將分別從裝有血清中的CRP與純CRP溶液的瓶裡各取出100μl之量;濃度從0.1~30(0.1,1,5,10,20,30) mg/l做為檢測樣品加入500μl試劑中並放入混頻交流磁導率計裡量測,藉由歸一化混頻交流磁導率χac減量之方法對不同濃度之血清中的CRP與純CRP溶液所作出曲線,並以兩曲線做比較來驗證磁性標記混頻交流磁導率計對量測帶有顏色的樣品,其準確性並不受影響。
初步的實驗結果顯示,磁性標記混頻交流磁導率量測法其敏感度確實具有與目前的酵素連結免疫吸附分析法相匹敵,但準確性絕對可以超越它。未來若能充分運用將會是非常具有前瞻性的。
